Human SARS-CoV-2 Spike specific single B-cells are FACS sorted. After amplification the variable regions are cloned into Heavy and Light pFUSEss plasmids. The antibody is brought to expression by co-transfecting 293T cells and subsequently affinity purified and concentrated.

SARS-CoV-2 Spike antigen specific binding is determined using ELISA: Secto (Sino Biological; 40589-V08H8) positive, S1 (Sino Biological; 40591-V08H3) positive and RBD (Sino Biological; 40592-V08H) positive. Antibody showed high affinity for RBD as determined by ELISA. Binding to SARS-CoV2 was also verified on protein array and no cross-reactivity to hCoV-OC43, hCoV-NL63, hCoV-HKU1, hCoV-229E was detected. Binding to SARS-CoV2 variants was also verified on protein array and reactivity to WT, Delta, Omicron BA.1, Omicron BA.5 was detected.
