Training in Coronavirus reverse genetics to adapt novel techniques (ISA method) to large RNA viruses and compare different technologies under BSL-3 conditions

SARS-CoV-2 diagnostic training, including differential diagnostics of human Coronaviruses and other respiratory pathogens, incl. BSL-2 and BLS-3 conditions.

Institut Pasteur Paris - Access to BSL 3 containment facility. The access will comprise a detailed theoretical and subject to availability practical application of the biosafety and biosecurity management at Institut Pasteur. Among others, establishment and validation of suitable decontamination methods for waist, rooms and material, as well as documentation and information management will be addressed.

In this 3-day course the basic principles of the protein microarray will be addressed by a combination of lectures, demonstration and hands-on training. The protein microarray technique can be used for HA antibody profiling for group level assessment of exposures to influenza viruses in small sample volumes from humans and animals (birds, horses, pigs, bats, carnivores a.o.). The training encompasses wet lab training but also the analysis and interpretation of assay results. Special attention will be given to specific issues of Influenza and flavivirus cross reactivity.

This five-day course includes two modules that can be attended separately. Participants will visit the Viroscience clinical virology lab, where they will perform all laboratory techniques including ELISA, Immunofluorescence and virus neutralization. Attention will be given to serological testing in cerebrospinal fluid, including the calculation of an antibody index.

In this 2-day course the basic principles of routine virus neutralization and ELISA assays will be addressed by a combination of lectures, demonstration and hands-on training. Special attention will be given to quality control and assay validation. Topics include: cell culture, biosafety procedures, preparation of sera and virus stocks, ELISA and microarray formats, different virus neutralization tests using pseudotyped viruses and wildtype viruses, immunofluorescence staining of cells, calculation of titers, assay validation and quality control.

Access to the BSL-3 facility including equipment and personal protective equipment, theoretical and practical training; preparation and evaluation of the BSL-3 work; BSL-3 virus collection as predefined within the project of the end user; personal support for performing specifically designed experiments; expert supervision.

In this 3-day course the basic principles of routine virus typing will be addressed by a combination of lectures, demonstration and hands-on training. Typing of enteroviruses will be used as an example, but the general procedures are applicable to typing of other viruses also. After this course participants should have sufficient knowledge to introduce routine typing of viruses in their own laboratory.
